【摘要】 目的研究水红花子的化学成分。方法利用各种色谱技术进行分离纯化,根据化合物的理化性质和波谱数据进行结构鉴定。结果分离得到4个化合物,分别为:3,3'-二甲基鞣花酸-4'-O-β-D-葡萄糖苷(3,3'-di- O-methylellagic acid-4'-O-β-D-glucopyranoside,Ⅰ)、3,3'-二甲基鞣花酸(3,3'-di-O-methylellagic acid,Ⅱ)、花旗松素(taxifolin,Ⅲ)、槲皮素(quercetin,Ⅳ)。结论化合物Ⅰ为首次从蓼科植物中分得,化合物Ⅱ为首次从该植物中分得

Abstract:ObjectiveTo study the chemical constituents of Fructus Polygoni Orientalis. MethodsVarious chromatographic techniques were used to separate and purify the constituents. Their structures were elucidated on the physico-chemical properties and spectral data. ResultsFour pounds were isolated from Fructus Polygoni Orientalis. Their structures were identified as 3,3'-di- O-methylellagic acid-4'-O-β-D-glucopyranoside(Ⅰ),3,3'-di-O-methylellagic acid(Ⅱ), taxifolin(Ⅲ), and quercetin(Ⅳ).Conclusionpound Ⅰ was isolated for the first time in polygonaceae, pound Ⅱ was obtained from polygonum orientale L. for the first time .

水红花子为蓼科植物红蓼Polygonum orientale L.的干燥成熟果实,具有健脾消食、化痞散结、清热明目、抑菌等作用,主治痞块腹胀、颈淋巴结核、消渴、胃痛、疝气和产后腹痛以及慢性肝炎、肝硬化腹水、脾肿大等〔1〕。药理实验表明其醋酸乙酯部分有较好的抑瘤效果。因此笔者对该药材醋酸乙酯部分的化学成分进行了研究,分离并鉴定了4个化合物。
1 材料与仪器
药材经辽宁中医药大学翟延君教授鉴定为红蓼Polygonum orientale L.的成熟果实。X4型显微熔点测定仪(温度计未校正);UV-265-FW型自记式分光光度计;IR200红外分光光度计;Burker-ARX-300 型核磁共振光谱仪;Angilent 1100 series ion trap mass spectrometer;LCQ型液相色谱-质谱联用仪。硅胶为青岛海洋化工厂生产。
2 方法与结果
2.1 提取与分离水红花子10 kg,粉碎, 75%乙醇多次提取,回收溶剂得乙醇浸膏。将浸膏混悬于水中,相继以石油醚、醋酸乙酯萃取数次,得石油醚部分、醋酸乙酯部分(215 g)和水溶部分。醋酸乙酯部分经反复硅胶柱层析(氯仿、甲醇梯度洗脱)结合制备薄层层析(石油醚-醋酸乙酯4∶1为展开剂)分离得到化合物Ⅰ~Ⅳ。
2.2 结构鉴定
2.2.1 化合物Ⅰ黄色粉末(丙酮-甲醇),mp.296~298℃。UVλmax(MeOH)nm:246.4,368.0。IR(KBr)cm-1:3 369(OH),1 748(C=O),1 609,1 578,1 488,1 440,1 415,1 363,1 247,1 195,1 174,1 088,984,915,757。1H-NMR(DMSO-d6)δ:7.82(1H,S,H-5'),7.55(1H,S, H-5),5.07(1H,d J=7Hz,H-1〃)3.22-3.71(m,糖的次甲基),4.08(3H,S,OCH3-3'),4.05(3H,S,OCH3-3)。13C-NMR(DMSO-d6)δ:158.5(C-7),158.5(C-7'),153.0(C-4'),151.7(C-4),141.9(C-3'),141.8(C-2'),141.1(C-2),140.3(C-3),114.3(C-1'),112.9(C-6'),112.3(C-5'),112.0(C-6),111.7(C-5),111.3(C-1), 61.8(OCH3-3'),61.1(OCH3-3),101.4(C-1〃),77.4(C-3〃),76.6(C-5〃),73.4(C-2〃),69.6 (C-4〃),60.6(C-6〃)。LC-MS m/z:515(M+Na+),331,316,299,237,228。其光谱数据与文献〔2〕对比,确定化合物Ⅰ为3,3'-二甲基鞣花酸-4'-O-β-D-葡萄糖苷(3,3'-di- O-methylellagic acid-4'-O-β-D-glucopyranoside)为首次从蓼科植物中分离得到。
