【摘要】 目的 用旋光度法替代酸碱滴定法测定盐酸麻黄碱滴鼻液的含量。方法 分别用旋光度法与酸碱滴定法测定盐酸麻黄碱滴鼻液的含量,并将其结果进行比较。结果 旋光度法在0.25%~1.5%(g/ml)浓度之间线性关系良好(r=0.9989),平均回收率(n = 9)为100.3%,RSD =0.42%;与酸碱滴定法所测得样品结果经统计学检验,差异无显著性。结论 旋光度法简便、快速、准确,可用于盐酸麻黄碱滴鼻液的含量测定。

Determination of ephedrine hydrochloride nasal drops by polarimetry

【Abstract】 Objective To determinate the content of ephedrine hydrochloride nasal drops by polarimetry instead of acid-base titrations. Methods Ephedrine hydrochloride nasal drops was quantified by polarimetry and acid-base titrations. The data were analyzed by t-test.Results The assay of polarimetry was linear over the range of 0.25%~1.5%(g/ml)(r=0.9989). The average recovery was 100.3%(n=9) with RSD =0.42%. The difference of the data obtained by polarimetry and acid-base titrations was not significant. Two methods were statistically equivalent for the determination of ephedrine hydrochloride nasal drops.Conclusion The polarimetry is easy, rapid and aurate for the determination of the content of ephedrine hydrochloride, and can be used as an alternative method of acid-base titrations to determinate ephedrine hydrochloride nasal drops.

《中国医院制剂规范》第2版〔1〕收载的“盐酸麻黄碱滴鼻液”处方组成为“盐酸麻黄碱10g,氯化钠6g,羟苯乙酯0.3g,蒸馏水加至1000ml”。其中含盐酸麻黄碱(C10H15ON.HCl)1%,含量测定方法采用酸碱滴定法,而该书收载的“复方呋喃西林滴鼻液”中盐酸麻黄碱的含量测定采用旋光度法,旋光度法较酸碱滴定法简便快速,准确可靠。为验证旋光度法测定“盐酸麻黄碱滴鼻液”含量的可靠性,本文将旋光度法与酸碱滴定法测定的结果进行了比较,现报告如下。
1 仪器与试药
AUY220电子分析天平,WZZ-1自动指示旋光仪;盐酸麻黄碱标准品,纯化水,其余试剂均为分析纯;1%盐酸麻黄碱滴鼻液(本院制剂)4批,批号060915-1、060915-2、060915-3、060915-4。
2 方法与结果
2.1 酸碱滴定法〔1〕 精密量取本品10ml,加乙醚10ml、酚酞指示液与麝香草酚蓝指示液各3滴,用氢氧化钠液(0.1mol/L)滴定,随滴随振摇,直至水层显持久的紫红色即得。每1ml氢氧化钠液(0.1mol/L)相当于20.17mg的C10H15ON.HCl。
2.2 旋光度法〔2〕 按《中国医院制剂规范》第2版附录“旋光度测定法”,用2dm测定管,温度20℃时,将样品置旋光仪测定管中,测定盐酸麻黄碱滴鼻液在589.4nm波长下的旋光度,直接读出旋光度数。
2.2.1 旋光度法标准曲线的制备〔3~4〕 精密称取经105℃干燥至恒重的盐酸麻黄碱标准品适量,加纯化水溶解,配成浓度约0.25%、0.4%、0.5%、0.6%、0.8%、1.0%、1.2%、1.5%(g/ml)的系列溶液,置旋光仪中,测得旋光度与浓度对应值见表1。以浓度C(g/100ml)为横坐标,旋光度α为纵坐标,用Microsoft Excel软件处理,绘制标准曲线(图1),经直线回归,求得回归方程。
